The principle of gas chromatography is based on a selective distribution of components between the stationary and mobile phases. The mobile phase is a carrier gas that flows past the stationary phase. The substances to be separated that are carried along by the carrier gas interact with the stationary phase. This interaction consists of a 'temporary' binding of the molecule to the stationary phase, which slows it down. Because each molecule often has its own specific binding strength or affinity for the stationary phase, different delays take place for substances with different affinities. This allows substances to be separated from each other. The substances that are delayed will therefore be detected later by the detector. This results in separated peaks in the output.

A gas chromatograph is mainly composed of:
